The control of bolting time in onion is an important approach for bulb and seed production, as onion plants which bolt do
not produce marketable bulbs and seed yields are dependent on floral induction. However, genetic and molecular studies about
bolting time in onion plants have not been examined yet to date. In order to understand the regulation of bolting time in
onion plants, we conducted the genetic crosses between late bolting-type cultivar (MOS8) and very early bolting-type cultivar
(Guikum). Segregation ratio of late to very early in F2 populations indicated that this lateness trait was determined by a dominant locus. We also analyzed protein profiles in onion
plants with different bolting time by a proteomics approach. Interestingly, a protein spot with significant similarities to
chromodomains of mammalian chromo-ATPase/helicase-DNA-binding 1 or heterochromatin protein 1, which is involved in the histone
modifications, was identified. Histone methyltransferase activity was also observed in onion plants. Taken together, these
results suggest that a genetic pathway may be involved in the modulation of bolting time in onion plants, though there is
no direct evidence that this protein spot obtained by proteomics is relevant to vernalization. 相似文献

Amino acid homology analysis predicted that rbmD, a putative glycosyltransferase from Streptomyces ribosidificus ATCC 21294, has the highest homology with neoD in neomycin biosynthesis. S. fradiae BS1, in which the production of neomycin was abolished, was generated by disruption of the neoD gene in the neomycin producer S. fradiae. The restoration of neomycin by self complementation suggested that there was no polar effect in the mutant. In addition,
S. fradiae BS6 was created with complementation by rbmD in S. fradiae BS1, and secondary metabolite analysis by ESI/MS, LC/MS and MS/MS showed the restoration of neomycin production in S. fradiae BS6. These gene inactivation and complementation studies suggested that, like neoD, rbmD functions as a 2-N-acetlyglucosaminyltransferase and demonstrated the potential for the generation of novel aminoglycoside antibiotics using

Caspase-11 is an inducible caspase involved in the regulation of cell death and inflammation. In the present study, we examined whether apoptosis signal-regulating kinase 1 (Ask1)-mediated signaling pathway is involved in the expression of caspase-11 induced by lipopolysaccharide (LPS). We found that the induction of caspase-11 was suppressed by the inhibitors of NADPH oxidase (Nox) or knockdown of Nox4 that acts downstream of toll-like receptor 4 and generates Ask1-activating reactive oxygen species. Overexpression of dominant negative tumor necrosis factor receptor associate factor 6 also suppressed the induction of caspase-11. Importantly, knockdown or dominant negative form of Ask1 suppressed the induction of caspase-11 following LPS stimulation. Taken together, our results show that Ask1 regulates the expression of caspase-11 following LPS stimulation. 相似文献

The hippocampus is associated with learning and memory function and shows neurochemical changes in aging processes. Calbindin
D-28k (CB) binds calcium ion with a fast association rate. We examined age-related changes in CB immunoreactivity and its
protein level in the gerbil hippocampus during normal aging. In the hippocampal CA1 region (CA1) and CA2, CB immunoreaction
was found in some neurons in the stratum pyramidale (SP) at postnatal month 1 (PM 1). CB immunoreactivity in neurons was markedly
increased at PM 3. Thereafter, CB immunoreactivity was decreased with time: CB-immunoreactive (+) neurons were fewest at PM 24. In the CA3, a few CB+ neurons were found only in the SP at PM 1 and in the stratum radiatum at PM 18 and 24. In addition, mossy fibers were stained
with CB at PM 1. CB immunoreactivity in mossy fibers was markedly increased at PM 3, thereafter it was decreased with time.
In the dentate gyrus, many granule cells (GC) in the granule cell layer were stained with CB at PM 1. CB immunoreactivity
in GC was markedly increased at PM 3, thereafter CB immunoreactivity was decreased with time. In Western blot analysis, CB
protein level in the gerbil hippocampus was highest at PM 3, thereafter CB protein levels were decreased with time. This result

Oxidative stress is one of predisposing factors to age-related neurodegeneration in the brain. In particular, thiol-containing
groups are susceptible to oxidative stress, which induces the formation of the disulfide bond and/or hyperoxidized form of
thiol-containing proteins. We observed the protein thiol levels in the hippocampal homogenates and also investigated changes
in hyperoxidized form of peroxiredoxin (Prx–SO3) immunoreactivity and proteins levels in the gerbil hippocampal subregions during normal aging. Levels of total thiol, non-protein
thiol, and protein thiol were decreased in the hippocampal homogenates with age. At post-natal month 1 (PM 1), pyramidal and
non-pyramidal cells in the hippocampal CA1 region (CA1) showed Prx–SO3 immunoreactivity. Prx–SO3 immunoreactivity in the cells was decreased by PM 12, thereafter, Prx–SO3 immunoreactivity in the cells increased again with age. In the CA2/3, Prx–SO3 immunoreactivity in pyramidal cells was not significantly changed; however, the immunoreactivity in pyramidal cells was very
low at PM 12. Prx–SO3 immunoreactivity in the dentate gyrus (DG) was distinctly changed during aging. At PM 1, Prx–SO3 immunoreactivity in granule and polymorphic cells was weak and strong, respectively. The immunoreactivity in the neurons
was decreased with age, not shown in any neurons at PM 12. Thereafter, Prx–SO3 immunoreactivity increased again with age. In addition, Prx–SO3 protein level in the hippocampus was lowest at PM 12. These results suggest that thiol-containing proteins are changed during

High- and low-methoxyl pectins were treated with pectin methylesterase (PME) and the functional properties of the resulting pectin gels were characterized. The degree of esterification of high- and low-methoxyl pectins decreased from 74.5% to 6.3% and 40.0% to 6.5%, respectively while not changing their molecular weight. Also, the addition of glucono-delta-lactone (GDL) dramatically affected the gel strength and the pH reduction by the GDL led to the increased syneresis of the pectin gels, which was also observed in the PME-treated samples. When flavor compounds were incorporated into the pectin gels, the flavor release from the gels increased with decreasing the degree of esterification due to increased hydrophilic properties. 相似文献
